Researchers at the National Institutes of Health and the University of Oxford have announced a major breakthrough in the decades-long quest for a universal flu vaccine, publishing results from a Phase 2 clinical trial showing 89% efficacy against multiple influenza strains.
Unlike current flu vaccines that target the rapidly mutating head of the hemagglutinin protein, the new vaccine — dubbed FluVax-U — targets the stalk region, which remains consistent across influenza A and B strains. This means one shot could provide protection for multiple years rather than requiring annual reformulation.
"This is the holy grail of influenza research," said Dr. Sarah Chen, lead researcher at the NIH's Vaccine Research Center. "We've shown that by redirecting the immune system's focus to the conserved stalk region, we can generate broadly neutralizing antibodies that work against seasonal flu, pandemic strains, and even avian influenza."
The 15-year study, published in the journal Nature Medicine, followed 3,200 participants across the United States and United Kingdom. The vaccine demonstrated 89% efficacy against influenza A (H1N1 and H3N2 subtypes) and 76% efficacy against influenza B strains over two flu seasons — far surpassing the 40-60% efficacy typical of seasonal flu shots.
The research team is now planning a Phase 3 trial with 25,000 participants, aiming for FDA approval by 2027. If successful, a universal flu vaccine could prevent an estimated 290,000 to 650,000 respiratory deaths globally each year.
Public health experts are hailing the development as potentially transformative. "Influenza has been one of the most persistent threats to global health," said Dr. Anthony Fauci, former director of the NIAID. "A universal vaccine would be one of the greatest public health achievements of the 21st century."
